Pareitropone, the most effective tropoloisoquinoline alkaloid against the leukemia P388 cell line (IC50 = 2.7 nM), is an attractive synthetic target because of its unique structure of annulated tropone and isoquinoline moiety as well as potent cytotoxicity. We have reported total synthesis of pareitropone and based on it, also tried various analogs. However, isoquinoline formation was highly depends on substrate, so we newly revised the synthetic approach in convergent manner. With modified synthetic methods, pareitropone was successfully synthesized from commercially available 8-bromo-6,7-isoquinolines in much reduced step but better yield. |
