We previously demonstrated the target-specific delivery of hyaluronic acid (HA) derivatives to the liver by real-time bioimaging using quantum dots. In this work, HA – interferon α (IFNα) conjugate was developed for target specific systemic treatment of liver diseases, especially Hepatitis C Virus (HCV) infection. Aldehyde modified HA was synthesized and conjugated with N-terminal amine group of IFNα. After confirmation of in vitro anti-proliferation activity in Daudi cells, target specific systemic delivery of HA – IFNα conjugate was carried out with a pharmacokinetic analysis. In vivo antiviral activity of HA – IFNα conjugate was confirmed from the elevated expression levels of 2′,5′-oligoadenylate synthetase 1 (OAS 1) which is an essesntial anti-viral protein induced by IFNα. The target specific HA – IFNα conjugate will be discussed for further clinical applications as a once a week injection dosage form for the treatment of HCV infection. |
