제목 Permeability and biocompatibility of Ba-Alginate encapsulated hepatocytes with gelatin and poly (vinyl alcohol)
초록 [Introduction]
Encapsulation of microorganisms, animal and plant cells is an interesting method for cell culture with high density or for continuous preparation of metabolic products (hormones , secretory proteins or extracellular enzymes) pharmaceutical , medical , food , and other industries.
In this study, the permeability of microcapsules was investigated using proteins with different molecular weight, such as lysozyme, ovalbumine, globulin, albumin, acylase I and immmunoglobulin G.
[Experimental]
FITC-labeled proteins were prepared by incubating purified protein with fluorescein isothiocyanate in sodium bicarbonate buffer (200mM), PH 9.0, for a period of 20 h at 4℃. Unconjugated FITC was removed by dialysis in PBS.
FITC labeled proteins were dissolved in a PBS at a certain concentration, the solution was then mixed with an aqueous solution containing PVA and BaCl2. The mixed solution was dropped into a sodium alginate solution and stirred by a magnetic bar. The microcapsules were washed and incubated in culture dishes. FITC labeled protein released from the core part of the capsule was measured using a kinetic microplate reader.
[Results and Discussion]
Molecular Weight Cut Off (MWCO) is one of the important parameters when evaluating immunoisolation of the capsules. It is defined as the lowest molecular weight of the substance that cannot permeate through the membrane of the capsules.
