A Tumor Microenvironment-Specific Polymeric Nanoassembly Harnessing ROS-Triggered Cleavage of Diselenide Linkages |

Elevated levels of reactive oxygen species (ROS) are considered as one of the important tumor microenvironmental cues since they exert the critical roles in tumor progression. Thereby, ROS-responsive drug carriers have drawn much attention with an expectation to enable the site-specific drug release when recognizing abnormally high levels of ROS. We developed a novel polymeric nanoassembly based on biocompatible poly (ethylene glycol) and selenol-containing polypeptide derivatives to form ROS-cleavable diselenide crosslink. The drug-encapsulated nanoassembly went through ROS-mediated destabilization and facilitated drug release since the diselenide crosslink was converted to hydrophilic selenic acid in response to hydrogen peroxide, one of the major ROS in tumor tissues. Owing to these features, the nanoassembly improved intratumoural accumulation of the drug and further overall therapeutic efficacy, suggesting their high potency as an efficient anticancer drug carrier. |
