제목 Polymeric NONOates-Templated Mineralized Nanoparticles for Intracellular Nitric Oxide Delivery
초록 Nitric oxide (NO) has been recognized as an important molecule in controlling apoptosis of cancer cells. In recent years, mineralized nanoparticles based on block copolymer templates have shown efficient loading potential of therapeutics and delivery ability to target disease sites. In this study, we aim to develop a NONOates-loaded mineralized nanoparticle (NONOates-MNP) that can intracellularly release nitric oxide (NO). The NONOates-MNP was prepared by CaCO3 mineralization in the presence of an NONOates-incorporated poly(ethylene glycol-b-polyethyleneimine) (PEG-b-PEI) block copolymer. At physiological pH, the CaCO3 core phase of the nanoparticle would effectively stabilize NONOates. In contrast, at an endosomal acidic pH, NONOates would be exposed to an aqueous environment by CaCO3 dissolution, thereby generating nitric oxide (NO) molecules at the cytoplasm. This NONOates-MNP can be used as a novel intracellular NO-releasing nanocarrier for cancer chemotherapy.
