BSA and trypsin can adsorb on PLA homopolymer because of hydrophobic interactions. The presence of PEG segments on the surface of microsphere should reduce the adsorption and denaturation of protein, and so improve the stability of entrapped drug and increase the release yields. The aim of this study was to prepare BSA loaded PLGA microspheres with a reduced initial burst in the in vitro release profile, by addition of new biodegradable polymer. PEG/PLGA multiblock copolymer with different PEG/PLA ratios was synthesized by ring-opening polymerization. Structure and physical properties of multiblock copolymer were determined by NMR, GPC, XRD, and DSC. The preparation of microparticles was based on W/O/W double emulsion method. The morphology and size distribution were viewed with SEM. The amount of BSA released and loaded was determined by the Lowry-Peterson procedure. |
