Dual Growth Factor-Immobilized PCL Microspheres as a Bioactive Bulking Agent for the Treatment of Fecal Incontinence |
초록 |
Fecal incontinence caused by damage or weakness of the anal sphincter is a socially disabling disorder that profoundly impacts on all aspects of quality of life. Although injectable bulking agents are considered as a minimally invasive treatment, its low efficacy to treat the fecal incontinence caused by the injection volume decrement (microsphere migration and resorption) still remained as a clinical challenge. In this study, we prepared dual growth factor (bFGF and NGF)-loaded polycaprolactone microspheres as a bioactive injectable bulking agent which may promote the function of sphincter muscle around anus as well as maintain the initial injection volume, and thus improve the cure rate of the fecal incontinence. The growth factors were immobilized onto the PCL microspheres via heparin immobilization. The growth factor release profile, and differentiation /maturation potential into the target tissue of muscle-derived progenitor cells on the microspheres were investigated. |
