Restoration of the damaged endothelium by rapid reendothelialization reduces the restenosis and thrombosis in the stent treatment for cardiovascular diseases. It is known that outgrowth endothelial cells (OECs) play a key role in advancing reendothelialization of injured vessel. In this study, we produced the OEC capturing stent which is immobilized by CD146 antibodies. CD146 is a transmembrane glycoprotein that is expressed in the whole human endothelium and also expressed in OEC. Additionally, we employed silicone nanofilament (SiNf) on metal surface for increasing the immobilized amount of anti-CD146 Abs. SiNf-modified surface was evaluated by FE-SEM, AFM, ATR-FTIR, and XPS. The amount of anti-CD146 Ab immobilized on SiNf significantly increased compared to control. Anti-CD146 Ab sample captured more OEC than bare and anti-CD34 Ab under in vitro perfusion. This approach will be a fascinating way for accelerating the re-endothelialization of a vascular stent. |
