Medium-chain α,ω-alkanediols are extensively used for monomer composing several kinds of polymers in industry. We constructed α,ω-dodecanediol bioproducing process using cytochrome P450 monooxygenases (CYP) in E. coli. The E. coli cells over-expressing CYP153A33 from Marinobatcter aquaeolei and Nfa22290 from Nocardia farchnica IFM10152 with CamAB redox proteins and FadL transporter were examined for whole cell biotransformation, resepectivley. Co-expressing of fadL with CYP153A facilitated the uptake of dodecanol substrate through cell membrane, which resulted in 5-fold increase of α,ω-dodecanediol production. Further, we enhanced the diol production by heme precursor optimization. Finally, 178mg/L and 440 mg/L of α,ω-diols were produced by whole cell biotransformation of CYP153A and Nfa22290 from 20 mM of 1-dodecanol, respectively. |
