α, ω-dodecanediols are used as building block in the polymer industries. In this study, we report a strategy for biosynthesis of α, ω-dodecanediols via catalysis of CYP153A33 from M. aquaeoleiVT8 and Nfa22290 from N. farcinica. Redox protein CamA/B from Pseudomona putida and long-chain fatty acid transporter FadL were introduced in E. coli. In addition, the dependence of heme precursor and iron on dodecanediol synthesis was investigated. Finally, 0.88 mM and 1.7 mM of dodecanediol were produced via the biocatalysis of CYP153A33 and Nfa22290, respectively, from 1-dodecanol under the optimized condition. |
