Highly selective, sensitive, and multiple detection sensor have become a critical challenge for disease diagnosis. In this article, we describe a multi-analyte biosensor for the detection of cancer biomarkers based on plasmon response of single gold nanoparticles (AuNPs) and antibody-antigen binding interaction. To demonstrate the ability to detect and quantify cancer biomarkers: prostate speacific antigen (PSA), a-fetoprotein (AFP), Carcinoembryonic antigen (CEA), a novel and sensitive multiplex sensor was developed by site-selectively immobilizing plasmonic nanoparticles functionalized with each monoclonal antibody that recognize the target protein on hydrophilic-hydrophobic patterned glass slide. Using this sensing strategy, we find that the multi-analyte ability, high sensitivity and simple detection method based on Rayleigh scattering of individual gold nanoparticles, which make it an excellent application for versatile molecular diagnostics in both research and clinical fields. |
