제목 pH-Tunable Calcium Phosphate Coated Porous Nanocontainers by Enzyme-Mediated Surface Mineralization
초록 Mesoporous silica nanoparticles (Si-MP) have been characterized by their unique features such as stability, biocompatibility, controllable pore size, and readily modified surface functionalities. The large surface area and pore volume of Si-MP have offered the possibility of encapsulating and delivering large quantities of drug molecules. In this study, we describe the surface functionalization and the urease-mediated mineral coatings of Si-MPs. The pH-controlled dissolution kinetics of CaP coatings and its relationship with the pH-controlled drug release were performed. The CaP coating could hold the encapsulated drugs in extracellular condition, whereas it could be dissolved within intracellular endosomes to accelerate the drug release. In vitro cytotoxicities of Si-MPs and surface-mineralized Si-MPs for breast cancer MCF-7 cells were estimated. Intracellular localization of drug-loaded mineralized Si-MPs was confirmed using confocal laser scanning microscopy.
