Due to impeded blood flow and hypoxia, severe cardiac cell death occurs in myocardial infarction (MI). Mesenchymal stem cell (MSC) implantation has therapeutic potential for heart repair. However, reactive oxygen species (ROS) generated in the infarcted region by reperfusion of blood flow, remains as a critical hurdle for survival of engrafted MSCs. Herein, we report the utility of graphene oxide (GO) sheets to provide ECM and shield the implanted MSCs from ROS and thereby enhance the therapeutic efficacy of implanted MSCs. MSCs culture with GO sheets were injected after reperfusion in infarcted region. In spite of possible damage from ROS, GO sheets were capable of protection and attenuation of anoikis, thereby showed increased cell viability and angiogenesis. Heart tissue of GO-MSCs injected mice showed efficient cardiac repair with decreased infarcted area and fibrosis. This study demonstrates that GO sheets can effectively enhance the engraftment capability of implanted MSCs. |
