Myocardial Infarction Treatment using Cardiac Lineage-optimized and Paracrine-improved MSCs from Iron Oxide Nanoparticle-mediated Co-culture Platform |

Electrophysiological phenotype development and paracrine secretion of mesenchymal stem cells (MSCs) determine the therapeutic efficacy of MSCs for myocardial infarction (MI). Thus, co-culture of MSCs with cardiac cells has windowed cardiac priming of MSCs. Particularly, gap junctional crosstalk in co-culture play a major role in cardiac modification of MSCs. Here, we report iron oxide nanoparticles (IONPs) augment the expression of gap junction protein (Cx43) of H9C2 to promote gap junctional communication with MSCs in co-culture. MSCs co-cultured with IONP-harboring H9C2 (cMSCs) displayed higher levels of cardiac biomarkers and cardiac repair-favorable paracrine profile. Accordingly, cMSC injection into rat MI models improved animal survival and heart function compared with normal MSC injection. Present study highlights a novel application of IONPs in developing intercellular gap junctions, and proposes a generation cMSCs that exceed the reparative potential of conventional MSCs. |
