A fluorescent probe(1) for detection of hydrogen sulfide was designed and synthesised based on the amino-terminal copper and nickel (ATCUN) binding motif and 4-Nitro-2,1,3-benzoxadiazole (NBD) fluorophore. NBD labeled tripeptide (Dap-Gly-His) formed the ensemble with Cu2+(1-Cu2+), which had turn-off response. The addition of hydrogen sulfide instantly removed Cu2+ and showed turn on response in 100% aqueous buffer solution. Among the bio-thiols, the ensemble (1-Cu2+) sensitively and selectively detected a wide range concentration of H2S ranged 2 μM to 750 μM in pure aqueous solutions by a fluorescent change. The response time for H2S was 30 min which is the removal of Cu2+ from the ensemble 1−Cu2+. Moreover, the cell image study was optimized by tuning the ATCUN binding motif sequence of the ensemble. It demonstrated that the probe can be employed to detect hydrogen sulfide in live cells. |
