In this research, we have created a new method using an integrated microfluidic device for efficient microalgae lipid analysis. We performed multiprocess including microalgae culture, lipid production and extraction on a device. This device is composed of cell growth chamber and connecting channel filled with micropillar array between growth chamber and output reservoir for efficient cell lysis and lipid extraction. Microalgae cells were grown for 4 days and lipid accumulation was induced by supply of nitrogen-deficient medium into microfluidic cell growth chamber. After that, cell lysis and lipid extraction were performed by flowing various solvents such as 70% ethanol and isopropanol at 60℃. The lipid was analyzed by TLC and gas chromatography. We confirmed the lipid extract from integrated microfluidic device contains neutral lipid (TAG) and fatty acid content is comparable to that from conventional Bligh-Dyer method. |
