ω-Transaminase (ω-TA) is an important enzyme for the synthesis of chiral amines or α-amino acids by transfer of an amino group from primary amines to α-carbonyl compounds. There are two approaches to prepare enantiopure amines using ω-TA. (i) Kinetic resolution of a racemic amine or (ii) asymmetric synthesis starting from a ketone. However, despite the availability of ω-TAs, inhibitions by substrate and product have been a challenge to use the ω-TA reaction for industrial processes. Here, we report the ω-TA devoid of substrate and product inhibitions and demonstrate the efficient kinetic resolution of racemic amine without product removal. |
