Increasing the soluble production of recombinant therapeutic proteins with an in situ-removable fusion partner in a cell-free synthesis system |

Bypassing the multiple steps of gene cloning and cell cultivation, cell-free protein synthesis provides a rapid platform for the production of recombinant proteins. However improving the yield of soluble proteins often becomes an important issue during cell-free production of recombinant proteins. In this study, as a tactic to increase the soluble expression of therapeutic recombinant proteins in a cell-free synthesis system, we used ubiquitin as an in situ-removable fusion partner. Upon the fusion with the ubiquitin sequence by PCR, we were able to increase the soluble production of proteins that are otherwise showed low productivity and solubility. Through the inclusion of a deubiquitinase in the reaction mixture, we were also able to remove the ubiquitin sequence during translation process, which allowed accumulation of target proteins in their native amino acid sequences. The presented method could be used as a facile route to prepare soluble proteins for various applications. |
