We present a polymeric prodrug of cinnamaldehyde, poly(cinnamaldehyde β-amino ester)-co-poly(ethylene glycol)(PCAEG), which causes ROS(reactive oxygen species)-mediated apoptotic cell death. PCAEG incorporates covalently cinnamaldehyde in the hydrophobic backbone through acetal linkage. It was designed to have two pH-responsive moieties in its backbone, protonable amine groups and acid-cleavable acetal linkages. The amphiphilic nature of PCAEG allows them to self-assemble to form micelles and encapsulate hydrophobic anticancer drugs. At the acidic tumor site, dual pH-sensitive PCAEG dissociates to liberate the drug payload and then release cinnamaldehyde. PCAEG micelles showed a pH-dependent micellization/ demicellization behavior and cinnamadlehyde release kinetics. Cell killing ability of PCAEG was evaluated by confocal microscopy and flow cytometry. The micelles also showed synergistic effects of PCAEG with payload on the induction of apoptosis. |
