Microbubbles (MBs) are widely used as ultrasound contrast agent and hasbeen actively investigated as drug carriers. However, the instability of MBs isa bottleneck in ultrasound diagnosis and drug delivery. The exosome, a type ofextracellular vesicles released from cells, has a stable membrane. In thisstudy, exosome-embedded microbubbles (Exo-MBs) were developed to overcome thelow stability of MBs by embedding the exosome membrane proteins into MBs. As aresult, the stability of Exo-MBs is improved over the conventional MBs.Therefore, Exo-MBs showed long lasting ultrasound signal compared to SonoVueⓇ. Also, chlorin e6-loadedExo-MBs (Ce6-Exo-MBs) are self-assembled into chlorin e6-loadedexosome-embedded liposomes (Exo-Lipos) and locally extravasate by externalultrasound. The Ce6-Exo-Lipos showed favorable accumulation properties for thetarget cells. They showed excellent therapeutic efficacy by photodynamictherapy as well as immunotherapy triggered by exosomal proteins. |
