In-situ monitoring of polymorph transformations of Active pharmaceutical ingredients (API’s) in Anti-solvent crystallization |
초록 |
Screening of API polymorphs is an essential component of drug development and improvement. From preclinical development through manufacture of the final drug product, the identification, selection and understanding of the solid forms has an enormous practical and commercial impact on drug development. In-situ monitoring is implemented to understand and control the crystallization of an active pharmaceutical ingredient in development. This necessitates optimizing the experimental conditions and control parameters using the appropriate number of precisely designed experiments to address the stage of development of the various forms of the drug substance under study. The transformation of form I to form II of CHS was studied by seeded and unseeded crystallizations in formic acid-isopropanol system. Initial solution concentration, seeding amount and time etc. were considered. |
