Extracellular matrix loaded moldable and injectable alginate-cellulose β-TCP beads for accelerated bone regeneration |
초록 |
Extracellular matrix (ECM) is a 3d frame of proteins, growth factors and macromolecules playing significant role in tissue regeneration. ECM obtained from allogeneic sources upon decellularization thus bearing a great hope in tissue engineering field. Even after decellularization, we detected optimum amounts of native bio functional proteins and macromolecules into ECM derived from porcine liver. The ECM was utilized with natural polymers sodium alginate and tempo-oxidized nanocelluloase in combination with bioactive calcium phosphate βTCP in order to fabricate beads for orthopedic application. Three groups of beads were developed without ECM- βTCP (AC), with only βTCP (TAC) and with both βTCP and ECM (ETAC). It was observed that cellular affinity and biocompatibility of beads were interestingly increased after addition of ECM. Densely cellular assemble surrounding the beads (ETAC) were visualized during five days of co-culturing with cells. After implantation of beads into rabbit femur, ETAC facilitated regeneration of more uniform cortical and cancellous bones than those of other two groups AC, and TAC. Al though the regeneration capacity of beads TAC was also notable, the healing speed and quality were quite faster with beads ETAC that indicated the synergizing action of ECM and βTCP for accelerated bone remodeling. Moreover mold-ability and inject-ability also rendered advantages to the beads for clinical application. |
