The Influence of Hybrid Scaffolds on the Inflammatory Reaction |

This study was designed to investigate the effect of demineralized bone particle (DBP) and porcine small intestinal submucosa (SIS) as a natural material on the local inflammatory reaction of Poly(D,L-Lactic-Co-Glycolic Acid) (PLGA) in vitro and in vivo. In this study, PLGA/20% of DBP/0, 10, 20, 40 and 80% of SIS scaffolds were evaluated using an NIH/3T3 cells were seeded into PLGA/DBP/SIS scaffolds. After 1, 2 and 3 day, proliferation and viability were measured by MTT assay. The morphology of cellular adhesion were observed by scanning electron microscope (SEM). The in vivo response of PLGA/DBP/SIS film were compared with that of PLGA film implanted subcutaneously; the local inflammatory response was observed according to histology. PLGA/DBP/SIS blends appear to be a promising approach for tissue engineering applications due to excellent biocompatibility and anti-inflammatory effects. |
