Mitochondria targeting doxorubicin delivery with bioreducible AB2 miktoarm polymeric carrier enhance therapeutic efficacy and survival rate in vivo |
초록 |
Recently intracellular targeting therapeutics delivery has attracted great attention in order to get enhanced therapeutic efficacy and reduced side effects. For mitochondria targeting doxorubicin (DOX) delivery, DOX was chemically conjugated with a bioreducible AB2 miktoarm polymer, which is composed of poly(ethylene glycol) (A) and poly(benzyl-l-aspartate) (B) blocks. Triphenylphosphorinum (TPP) as mitochondria targeting moiety was introduced physically or chemically to the self-assembled DOX-conjugated AB2 miktoarm nanoparticles (NPs). The size of the DOX and TPP loaded NPs were 100-200 nm in diameter. In vitro studies in KB and MDCK cells showed that TPP loaded NPs enhanced mitochondrial uptake ratio of DOX compared to free DOX. In vivo studies also demonstrated significant tumor size reduction with longer survival profile for TPP loaded NPs. |
