Synthesis of dual-responsive micelles based on Diels-Alder click reaction for effective doxorubicin delivery |

A dual-responsive core cross-linked micelles based on amphiphilic block copolymer of poly(oligo(ethylene glycol) methyl ether methacrylate)-block-poly(styrene-alt-maleic anhydride) (POEGMA-b-PSMA) was prepared. The POEGMA-b-PSMA triblock copolymers were synthesized by the RAFT polymerization and subsequently functionalized with furfuryl amine. Diels–Alder reaction was carried out to crosslink the hydrophobic core at 60 °C. The drug loading content and efficiency were estimated to be 18.0 (wt%) and 90%, respectively. TEM images showed spherical shapes of DOX loaded cross-linked micelles with an average hydrodynamic diameter of 70 nm. The micelles displayed a good stability under physiological condition but disassociated immediately under reductive acidic environment of tumors. Finally, the in vitro drug release showed that DOX release from micelles was only 11 % at pH 7.4, whereas 62% DOX was released at pH 5.0 and 10 mM glutathione. |
