In vitro study of large-diameter vascular grafts under pulsatile shear stress using elastic poly(L-lactide-co-ε-caprolactone) scaffolds |
초록 |
Although there has been an increasing demand for prosthetic vascular graft, no viable synthetic graft was developed yet. Related problems are growth potential, poor mechanical property, biocompatibility, and limited patency. Especially, in case of large-diameter vessel exposed to a high flow condition, appropriate mechanical properties are essential due to the safety issues. In this study, very elastic, biodegradable poly(L-lactide-co-ε-caprolactone)(PLCL) was used as a scaffold material and three dimensional tubular scaffolds were fabricated by an extrusion-particulate leaching method. The morphology of scaffolds was observed using a scanning electron microscope. The mechanical properties and elasticity of the scaffolds were investigated in terms of tensile tests and recovery measurements. We hypothesized that bio-mimetic culture conditions with applied pulsatile mechanical strain can accelerate vascular tissue ingrowth and maintain its phenotype similar to native tissue. To investigate this hypothesis, smooth muscle cells (SMCs) isolated from rabbit aortas were cultured in 3-dimensional tubular scaffolds for 7 weeks under pulsatile shear stress, compared with static culture.
