제목 Temperature-triggered Self-assembly of Elastin-resilin Fusion Proteins for Advanced Drug Delivery
초록 Stimuli-responsive polypeptide-based block copolymers that can be self-assembled into various nanostructures have been studied for drug delivery applications. We report herein molecular design, expression, purification and characterization of elastin-resilin fusion proteins and their temperature-triggered self-assembly. Temperature-responsive fusion proteins composed of elastin-based polypeptides (EBPs) and resilin-like polypeptides (RLPs) were genetically engineered, purified by inverse transition cycling, and the thermally induced self-assembled nanostructures were characterized by thermal profiling, dynamic light scattering and transmission electron microscopy. These self-assembled nanostructures were reversibly formed in response to the change in temperature depending on the temperature-responsiveness of EBP and RLP fusion proteins. The EBP and RLP fusion proteins with tunable thermo-responsive properties would make them attractive for advanced drug delivery systems.
