Characterization of Binding Affinity of Growth Factor Receptor Targeting Polypeptides by Enzyme-linked Immunosorbent Assay |

Binding affinity of a targeting ligand against a growth factor receptor (GFR) in the cell matters because it determines whether or not intracellular signaling could proceed. In this study, GFR binding fusion polypeptides composed of thermally responsive elastin-based polypeptide (EBP) block copolymers and GFR targeting peptides were genetically engineered, expressed, purified and their binding affinity against the GFR was characterized by enzyme-linked immunosorbent assay (ELISA). The GFR binding fusion polypeptides with hydrophilic EBP blocks showed soluble unimer form while those with various EBP diblock copolymers representing temperature-triggered core-shell micellar structures with multivalent GFR targeting peptides. Depending on spatial display of zGFR targeting peptides, their binding affinity to GFRs was controlled. This work would suggest how binding affinity of the GFR targeting peptides could be controlled based on multivalency. |
