Tissue engineering for cartilage regeneration using a Hyaluronic acid/ PLGA scaffold : in vivo test |

PLGA and hyaluronic acid(HA) has been widely used as biocompatible scaffolds materials to regenerate tissue. In this study we prepared HA/PLGA scaffolds by emulsion freeze-drying method. Chondrocytes were isolated from the articular cartilage of rabbit and seeded on the HA-PLGA scaffolds. After 2weeks of cell seeding, we implanted HA/PLGA scaffolds on the back of nude mice. Morphological observation, histology, biological assay for collagen and sGAG, and PCR were performed at each time point 1, 2, 3 and 6 weeks. Specific mRNA, B-actin and collagen type II, for chondrocyte and cell distribution expressed highly in HA/PLGA scaffold while dedifferentiation marker. Although collagen, sGAG, collagen type II and aggrecan were greater than HA-PLGA scaffolds. Therefore, this study suggests that HA/PLGA scaffold may serve as a potential cell delivery vehicle and a structural basis for in vivo tissue engineered articular cartilage.This work was supported by SCRC(SC4110). |
