Fibroblast-derived extracellular matrix-covered Co-Cr stents for endothelial progenitor cell delivery |

Rapid re-endothelialization at the inserted stent in atherosclerotic lesion may be effective to reduce or to prevent local restenosis and thrombosis. In this study, fibroblast-derived extracellular matrix (FDM) was fabricated on cobalt-chromium alloy (Co-Cr), resulting in a reservoir, for delivery of endothelial progenitor cell (EPC). Since EPC can be differentiated to endothelial cell (EC) in a suitable condition, like extracellular matrix (ECM), we established covalent bond formation between Co-Cr and FDM using carboxyl ended-self assembly monolayer (SAM) on Co-Cr surface. We assumed that covalently bounded FDM on Co-Cr is effective for EPC delivery even in the blood flow. Fibroblast cultured was decellularized by treating Triton X-100 and NH4OH, and the FDM structure was defined by immunostaining. In summary, we expect that FDM-bound Co-Cr surface may accelerate re-endothelialization as compare to bare metal surface. |
