A fluorescence probe (1) for H2S was synthesized based on the amino-terminal copper and nickel (ATCUN) binding motif and 4-Nitro-2,1,3-benzoxadiazole (NBD) fluorophore. NBD labeled tripeptide (Dap-Gly-His) formed the ensemble with Cu2+ for a fluorescent detection for H2S. The addition of H2S instantly removed Cu2+ from the ensemble (1−Cu2+) in pure aqueous solutions, resulting in a "turn-on" fluorescent response. Among the biothiols, the ensemble (1−Cu2+) sensitively and selectively detected a low concentration of H2S ranged from 10 µM to 100 µM in aqueous solutions by a fluorescent change. The response time for H2S was 30 min which is the removal of Cu2+ from the ensemble 1−Cu2+. We further turning the ATCUN binding motif sequence of the ensemble for the optimization of the detection of H2S in cells. |
