제목 Optimal n-butanol production from glucose and galactose based on carbon flux-associated redox rebalancing
초록 Although cellular redox state plays an important role for the production of chemicals and fuels by cell factories, it is difficult to achieve optimal redox rebalancing of engineered strains due to the sensitive responses of cellular physiology with regard to the redox state changes as well as carbon fluxes. Here, we demonstrated optimal rebalancing of the redox state enables maximizing cellular performance using n-butanol producing E. coli as a model system. This was achieved by tuning the expression level of yeast formate dehydrogenase based on UTR engineering. In results, flux of n-butanol was enhanced up to 35.4% and 44.6% from glucose and galactose, respectively. Interestingly, optimal production of n-butanol required different amounts of NADH depending on the substrate. Collectively, this work suggests that the advanced redox rebalancing should be considered for strain improvements.
