제목 | Increasing soluble expression of constitutive lysine decarboxylase using Consensus-driven Soluble Mutant Screening (ConsenSing) |
초록 | Although numerous studies had beenconducted to improve protein solubility using various approaches such ascomputational prediction of hot spots, addition of solubility tags, and modificationof surface charges, low correlation of computational and experimental data and tedioustrial-and-error process of those approaches hinder researchers moving on from commonstrategies like engineering promoter, expression conditions, and co-expressionwith chaperones. Therefore, we devised a system called ConsenSing (Consensus-driven Soluble Mutant Screening)to improve soluble expression of the constitutive lysine decarboxylase (ldcC)in E. coli by screening consensussequence based library with split GFP system. As a result, we have identifiedthree soluble mutants with seven, five, and eight mutations that show 10.8,5.5, 6.6-fold increase in solubility, respectively. |
