Microscale Programmed Hydrogel Designed for Biomimetic Intestinal Organoids |

The shrinkage or expansion of a hydrogel were widely used as sensitive scaffold, but limited prediction to volumetric changes in uncomplicated structure. In this study, we present that modeling of the shrinkage and expansion of responsive hydrogel using expansion ratio of hydrogel with structural ratio in a complicated hydrogel exposed to external stimuli. Then, we fabricated three-dimensional collagen villi scaffold using programed hydrogel reverse mold which can control dimeter of villi from 420 µm to 520 µm. As biomimetics, we cultured intestinal epithelial cells on three-dimensional collagen villi scaffold. In addition, to analyze the host-probiotics interaction in this hydrogel, co-cultured with Lactobacillus rhamnosus GG. Overall, these results will be highly useful for designing advanced tissue engineering scaffold that can enhance the quality of wide array of regenerated biomimetic tissues including three-dimensional villi. |
