A hypersensitive CaMKII assay using [γ-32P]ATP and a PSPD-gold binding peptide fused substrate |
초록 |
Protein interactions in biological systems are frequently controlled by protein kinases. Among them, Ca2+/calmodulin-dependent protein kinase (CaMKII) has attracted much attention because aberrant protein kinase activity has been implication in a variety of human diseases.Using the pspd gene encoding the 32-kDa putative spermidine synthase (PSPD) of Selnomonas ruminantium, a PSPD-gold binding peptide fused substrate was cloned and purified to give a highly selective substrate for CaMKII.Using [γ-32P]ATP the substrate was used for the highly sensitive detection of a phosphorylation catalyzed by CaMKII. |
