Diversification of protein-structure space is a major concern in protein engineering. Deletion mutagenesis can generate a protein sequence-structure space different from substitution mugenesis moediated space, but it has not been widely used in protein engineering because it causes protein misfolding. In this study, we demonstrated that such a problem can be overcome by co-introducing the stabilizing mutations together with deletional mutations. GFP was used as a model system, and various N-, C-, or loop deletion variants of GFP were sucessfully generated based on the devised approach. |
