Epidermal growth factor (EGF) is widely used in wound healing hydrogel research. In this study, Acrylate group was modified to EGF to cause a thiol-ene reaction between the acrylate group and the thiolated hyaluronic acid (HA-SH). HA-SH and methacrylated hyaluronic acid (HA-MA) were used as materials for hydrogel. As in the previous reaction, they were exposed to blue light in the presence of Riboflavin and then conjugated through a thiol-ene reaction. Modification of EGF was confirmed through Maldi-tof. Chemical analysis of hydrogels such as FT-IR and Raman represents that gels were successfully synthesized, and the strong improvement after gel formation was found through thermogravimetric analysis (TGA) and rheology. Gel degradation and EGF ELISA confirmed that the release of Acrylate-modified EGF was delayed. In addition, MTT assay showed high biocompatibility of hydrogel as an injectable use. In scratch assay, proliferation and migration of cells show wound healing ability of EGF conjugated hydrogel. As a result, Acrylated modified EGF, which is continuously released from hydrogel, is steadily involved in wound healing. |
