Development of dual scaffolds using multi channel granule and hydrogel for cartilage tissue regeneration. |

A Hyaluronic acid (HA)/gelatin (gel) hydrogel combined and extruded into a multi channel grandule creating a cartilage-bone dual scaffold then BMP-7 was applied to promote osteoinduction along with the slow releasing drug poly(lactic-co-glycolic acid) (PLGA). The physical properties such as porosity determined by mercury intrusionporosimetry, surface by scanning electron microscopy (SEM), chemical composition by Fourier transform infrared spectroscopy (FTIR) and crystallography by X-ray diffraction. An in vitro experiment was done with a MTT assay to study the cell viability of MC3T3-E1 preosteoblasts and ATDC5 prechondrocytes due to the effects of PLGA, cell proliferation and differentiation studied using a confocal fluorescent microscope. A polymerase chain reaction (PCR) is done to investigate the differentiation of chondrocytes with an in vitro sample when exposed to differentation media in comparison to a control sample. An ELISA test is done to investigate the levels of osteopontin (OPN), osteocalcin (OCN) and collagen I. An in vivo experiment was done by implanting the scaffold into the articular cartilage of a rabbit model, from the results of the HA-gel hydrogel and MCG combination with BMP-7 growth factor, the MCG boasts strong mechanical properties against the stress given from the compression given to the knee and the hydrogel's porosity and cell proliferating properties allowed differentation of substantial osteoblasts and chondrocytes along with cell to cell communications all of these between the MCG, HA-gel hydrogel and the cancellous bone region. These results show great promise in the study and use of dual scaffolds in the future. |
