Under shaking conditions, lower yields of microbial cellulose(MC) were reported as typical characteristics of Acetobacter xylinum ATCC 23769. To achieve satisfactory yield under shaking conditions, a novel scheme and a complex medium were developed for the production of microbial cellulose by A. xylinum. The optimum culture medium was determined to contain 0.5% peptone, 2.0% yeast extract, 0.5% glucose, 0.156% Na2HPO4, 0.18% KH2PO4, 50 ppm MgSO4, 2 ppm FeCl3, 0.5% citric acid, and 10 ml of ethanol per liter. Cellulose productivity achieved with this medium under static culture conditions was 0.446 for glucose which is much higher than reported. Cellulose productivity obtained under shaking conditions above 250 rpm was 0.576 which is still higher than that of static culture and much higher than those reported elsewhere for the static cultures. The kinetics of cell growth and cellulose production in the developed medium under static and shaking culture were also investigated.
