Conjugation of antibodies and gold nanoparticles has been applied for drug delivery, gene transfer, clinic diagnosis and environmental analysis. Here in, we fabricated a lateral flow immunoassay that is a general method for detection of infections. Different sized gold nanoparticles ranging from 34 to 138 nm were synthesized and then conjugated with antibodies (Hepatitis B). We focused on conjugation efficiency between antibodies and gold nanoparticles with different sizes. Conjugation was optimized at different pH values and concentrations of antibodies by UV-vis spectroscopy. The optimized pH and concentration were 7 and 3 μg, respectively. Among different sized gold nanoparticles, the 43 nm gold nanoparticle-based lateral flow immunoassay showed the greatest sensitive performance for detection of Hepatitis B virus. Our later flow immunoassay based on well-controlled gold nanoparticles could be applicable for many other biological and chemical analytes. |
