제목 GALACTOSYLATED CHITOSAN-graft-POLY(VINYL PYRROLIDONE) AS HEPATOCYTE-TARGETING DNA CARRIER
초록 Chitosan has been widely used in pharmaceutical research and in industry as a carrier for drug delivery and as biomedical materials for artificial skin and wound healing bandage applications due to its non-immunogenecity, good biocompatibility and non-toxicity. In previous studies, galactosylated chitosan (GC)-graft-dextran and GC-graft-poly(ethylene glycol) (PEG) were synthesized and checked as hepatocyte-targeting gene carriers. The complexes were stable and non-toxic. Also, the transfection of the complexes only occurred in the cells with asialoglycoprotein receptors (ASGPR), as a indication of receptor-mediated delivery of DNA into the hepatocyte. But transfection efficiency of the complexes was still low compared with that of lipofection. In this study, the GC-graft-PVP (GCPVP) was prepared and characterized. Also, the complex formation between GCPVP and DNA was checked for hepatocyte-targeting gene carrier.
