Encapsulation of Quantum dots with Mixture PEG-PCDA and PCDA-Herceptin conjugates for Targeted Cancer Imaging |

Polymer based nanoparticles are now widely using for drug delivery and target therapy. We modified the surface of QDs in solid dispersion method by PEG-PCDA & PCDA-Herceptin conjugates to make it water soluble & target specific. Upon UV-irradiation, the core of QDs in micelles was further stabilized by intra-micelles cross-linking between neighboring PCDA moieties. The QDs micelles were spherical with diameters around 130nm. Herceptin is a humanized IgG monoclonal antibody which targets the epidermal growth factor receptor-2 (HER-2). In this study we successfully make a conjugates with highly fluorescent non-toxic QDs (CdSe/ZnS). HER-2 positive expressed breast cancer cell SK-BR3 shows brighter image than HER-2 negative expressed breast cancer cell MDA-MB231 & KB cell (non containing HER-2 receptor) due to having herceptin with this conjugate. |
