제목 Microwell Culture of Neurospheres Enhances Human Neural Stem Cell Self-renewal and Differentiation
초록 Enhancing self-renewal and differentiation of human neural stem cell (hNSC) is of great importance for improving the therapeutic efficacy of treating neurodegenerative disorders. hNSCs proliferate and form neurospheres in undifferentiated status. However the conventional hNSC culture methods do not allow for precise control of microenvironments for neurosphere culture. Controlling the neurosphere microenvironments is involved in the regulation of hNSC differentiation. Here, we report neurosphere culture in the microwells (500um diameter) for enhancing hNSC self-renewal and differentiation. This was accomplished by forming controlled-size neurosphere in microwells. We found that control of neurosphere size enhanced self-renewal and differentiation to neuronal and glial lineage compared to conventional culture methods. Therefore, the approach using microwell could be useful for regulating hNSC behavior.This work was supported by the NRF (2010-0020409, 2013-Fostering Core Leaders of the Future Basic Science Program).
