제목 Controlled and Sustained Drug Delivery Through an In Situ Cross-linkable Micelle-conjugated Hydrogel For Cancer Treatment
초록 Chemotherapy has gained attention due to the improved efficacy and low side effects. Although nanocarriers and hydrogels have been studied as hydrophobic drug carriers, it is still challenging due to their low stability and hydrophilic nature. Here, we prepared the in situ crosslinked Tet-TA(Tetronic-tyramine) micelle/gelatin hydrogel composites via HRP/H2O2-mediated cross-linking for the sustained delivery of anticancer drugs. Paclitaxel (PTX) was successfully loaded (59.9%) into the Tet-TA micelles. PTX-loaded composites were simply formed via enzymatic crosslinking and the micelles were crosslinked to the hydrogel, increasing the stiffness (6.6→7.3 kPa). Moreover, in vitro sustained release of PTX (~47% in 28 days) was evaluated. The cell study showed drug efficacy (viability = 60.7→34.7%). In conclusion, we expect the in situ crosslinked micelle/hydrogel composite can be a promising lipophilic anti-cancer drug carrier.
