Inducing angiogenesis of human adipose-derived stem cells using pH-sensitive gold-iron nanoparticles |

Stem cell-based therapies for ischemic diseases have shown promising results which have several drawbacks such as low cell survival and engraftment after transplantation. Therapeutic efficacy of adipose derived stem cells (ADSCs) is majorly induced by angiogenic paracrine factors. We synthesized pH-sensitive gold-iron nanoparticles (AuFeNPs). The concentration of iron (Fe) ion is related to cellular hypoxia and secretion of angiogenic factors such as vascular endothelial growth factor and fibroblast growth factor. Fe ion, delivered to ADSCs using AuFeNPs, enhanced angiogenic factor secretion. Fe ion released from AuFeNPs in pH-sensitive manner. AuFeNPs showed increment in angiogenic factors secretion from hADSCs, compared to equal concentration of AuNPs treated hADSCs. Our novel nanoparticle used in this study as a carrier for pH-responsive intracellular metal ion delivery may open a new method for future therapy. |
