제목 Identification of a novel bacterial immunostimulatory protein and its application for the development of a vaccine adjuvant
초록 To minimize non-specific adverse immunological effects, an adjuvant should specifically enhance the immune response to the vaccine antigen with which it is co-administered. Therefore, developing a methodology that ensures the co-localization of adjuvant with vaccine antigen would be beneficial. To date, most vaccine adjuvant, comprised mainly of chemical substances, have been administered as a mixture with vaccine antigen. Hence, identification of a protein adjuvant that can be chemically linked with commonly used protein antigens can open a new paradigm for vaccine development. In this study, we have developed a new high-throughput approach called “EPSIA”, Expressed Protein Screen for Immune Activators and identified Vibrio cholerae phosphatidylserine decarboxylase (PSD) as a novel bacterial protein that activates host innate immunity. PSD stimulated murine macrophages to secrete proinflammatory cytokines via toll-like receptor 4 in a MyD88-dependent manner. PSD-induced proinflammatory response is mediated, in part, by its enzymatic activity. Most importantly, PSD also exhibited strong adjuvant activity inducing a Type 1 cell-mediated immune response against a co-administered inert antigen. These results reveal the versatility of the EPSIA technology for uncovering a new microbial stimulator of host innate immunity and further demonstrate the potential application of PSD as a vaccine adjuvant.
