제목 Transfection and intracellular trafficking properties of carbon dot-gold nanoparticle molecular assembly conjugated with PEI-pDNA
초록 To understand the mechanisms of polymeric gene delivery, monitoring of polyplex dissociation inside cell is one of the most important issues. Even though there are many reports about cellular uptake and endosomal escape, the study on the dissociation of polyplex has not been reported thoroughly. Herein, we developed a delivery system which can monitor the dissociation of polyplex, and deliver the pDNA effectively. The polyplex dissociation was monitored using carbon dot (CD) since its excellent optical properties and biocompatibility. The fluorescence of CD is quenched by Au nanoparticle when they interact with non-labeled pDNA since CD and Au surface is modified with highly cationic polymer, polyethylenimine (PEI). When the pDNA was dissociated, the fluorescence of CD recovered and this changes enabled the monitoring of dissociation. In addition, this system showed high transfection efficiency with high cell viability which is the prerequisite of the effective carrier.
