Non-ionic Multifunctional Nanoaggregates Simultaneous Delivery of Doxorubicin and Oligonucleotide by Cleavable Linkages |
초록 |
Cationic carriers containing oligonucleotide in current study an electrostatic interaction between cationic polymers and anionic oligonucleotide. However, cationic polymer complex are cytotoxic and oligonucleotide cannot be easily unloaded from the complex. In this study, doxorubicin and oligonucleotide were simultaneously conjugated to multi-armed-PEG with cleavable linkages and self-assembled into non-ionic nanoaggregates by hydrophobic interactions of doxorubicin. This conjugate spontaneously formed nano-sized core-shell strctures, which was confirmed by dynamic laser scattering and transmission electron Microscopy. Controlled release of doxorubicin and oligonucleotide were determined by measuring fluorescence intensities of librated molecules. Therefore, non-ionic oligonucleotide nanoaggregates containing high payload of doxorubicin are expected to be a multifunctional carrier system simultaneously aiming to gene therapy and chemotherapy for anti-cancer treatments. |
