In Vitro and In Vivo Bioactivity Following Intermittent Release of Osteotropic Molecules for Bone Regeneration |

For local bone regeneration without injections or oral dosing, a proper intermittent (IM) delivery system is needed. The association polymer system of cellulose acetate phthalate and Pluronic F-127 was used to make microspheres (MS) containing simvastatin (Sim) and/or Parathyroid Hormone (PTH) for local bone therapy. Drug-loaded and blank MS were alternately pressure-sintered to make 10 layer devices. In vitro effects of Sim and PTH were evaluated by constantly or intermittently exposing MC3T3-E1 osteoblastic cells to various Sim and PTH concentrations. Cultures were assayed for DNA content and AP activity. Implants were tested in a calvarial onlay model using male Sprague-Dawley rats. Devices examined were blank (no drug), sustained release, and IM implants. After up to four weeks of implantation, calvaria were harvested and prepared for decalcified histology. In addition to qualitative observations, thickness of newly formed woven bone was determined. |
