We report on the identification of a short linear motif peptide targeting A549 human lung adenocarcinoma through biopanning using a M13 phage library. In vitro and in vivo (subcutaneous and orthotropic tumor model) experiments demonstrate that the selected peptide show a highly selective binding affinity to A549. As the high level of fibrinogen is related to extrahepatic expression in lung and A549 cell response, interaction between the targeting peptide and immobilized fibrin was studied here. The excessive fibrin deposition induces epithelial -mesenchymal transition (EMT) which initiates tumor metastasis. Therefore, excessive deposited fibrin around A549 can be a new biomarker for tumor metastasis. Molecular dynamic calculation suggests that the short linear motif peptide specifically target an empty space of beta strand(B2) of fibrin, which is a cleft generated from the structural transformation of fibrinogen.
